Anotace	Allosteric modulators are known to modify the affinity of muscarinic receptors for orthosteric ligands but the sites of their attachment to receptors and the nature of the conformational change which they induce in receptors have been little clarified. Experiments on mutated receptors of M2 and M3 subtype have revealed that the third outer loop of the receptor plays important roles both in the high-affinity binding of gallamine and alcuroium and in the mechanism of their negative and positive allosteric effects
