We have synthesized a probe consisting of CdTe QDs conjugated with oligonucleotide to be used as a donor. Conjugation reaction between carboxylated QDs and aminated oligonucleotides has been performed via zero-length cross-linkers. Sample consisting of clinically-relevant PCR amplicons labelled with ROX (6-carboxyrhodamine) served as acceptor. Upon mutation-specific sample-probe hybridization an increase in fluorescence intensity at 610 nm (the emission spectra maximum of ROX) confirmed a proper function of FRET at a presence of detected mutation. The limit of detection evaluated for PCR amplified fragments of the sample is 4x10-9 M.
