Current study has been designed as a preclinical trial where superoxide dismutase 1 (SOD1) transgenic rats were used as a model for amyotrophic lateral sclerosis. An intrathecal application (5x105 cells) of human bone marrow mesenchymal stromal cells (MSCs) after disease manifestation led to ameliorated disease progression, significantly improved motor activity, and prolonged survival. For the first time, we reported that SOD1 rats have an abnormal disorganized PNNs structure around the spinal motor neurons and give different expression profiles of chondroitin sulfate proteoglycans (CSPGs) and corresponding genes. We reported that application of MSCs is a safe procedure having neuroprotective (showed by preserved PNNs structure accompanied by better survival of motor neurons, by remodelling the recipients’ pattern of gene expression) and immunomodulatory effects.
