    AnotaceThe acrosome reaction (AR) is a process of membrane fusion and lytic enzyme release, which enables sperm to penetrate the egg surroundings. Using super-resolution microscopy, we demonstrated that CD46 is relocalized during the acrosome reaction across the sperm head. This fact suggested that its role does not end with a loss of the acrosome vesicle and CD46 is probably a component of a large multiprotein complex on the sperm membrane that is responsible for sperm-egg interaction. Moreover, we showed that β1 integrins possess a similar position and behaviour as CD46 and they are present in the acrosomal area of the mouse sperm head and relocalized during AR. Thanks to STED and SIM super-resolution microscopy, we were able to distinguish a mutual position of these protein which was validated by co-localization analysis of super-resolution images. An association between CD46 and β1 integrin subunit was confirmed by proximity ligation assay. Moreover, we also defined a position of integrin α3 and α6 subunit in the sperm head and based on the obtained data they could be pairing with subunit β1, but probably occupy different sperm head compartments.
