We have applied Random amplified polymorphic DNA (RAPD) analysis to three newt species T. cristatus, T. dobrogicus and T. carnifex occurring in the Czech Republic and Slovakia to identify species-specific molecular markers, potentially useful for newt determination. Out of twenty randomly used RAPD primers, just one did not amplify any DNA fragment. Amplification reactions from the other 19 primers yielded 2 to 22 bands per primer. The total number of scored bands was 202. The average number of bands per primer and individual was 5.3. While the similarity indexes among individuals within populations (0.94 ů 0.99) and between populations within species (0.81 ů 0.98) were very high, between-species similarity was relatively low with the lowest value for T. carnifex and T. cristatus (0.49) and the highest value for T. dobrogicus and T. cristatus (0.53). Although nineteen primers amplified DNA fragments, only twelve generated clear highly reproducible species-specific bands.
