In this study we used scanning mutagenesis of the residues within the putative inner pore S6 region of TRPA1 channel with the aim of understanding the process of TRPA1 activation. We identify residues proline 949, asparagine 954 and glycine–(X)3-glycine motif as structural determinants of TRPA1 activation and deactivation and these findings contribute to our better understanding of molecular mechanisms of pain sensation
V této strukturně-funkční studii jsme se zaměřili na identifikaci klíčových aminokyselinových zbytků oblasti šesté transmembránové domény lidského TRPA1 kanálu, nezbytných pro jeho aktivaci. Za použití molekulárně-biologických a elektrofyziologických technik jsme ukázali, že prolin 949, asparagin 954 a motiv glycin–(X)3-glycin hrají zásadní úlohu v mechanizmech aktivace a deaktivace tohoto kanálu a tato zjištění přispívají k našemu hlubšímu pochopení vzniku bolestivých podnětů na nociceptivních neuronech
