Bacterial gene expression is controlled by sigma (σ) factors, that bind to RNA polymerase (RNAP), recognize specific promoter sequences in DNA, and mediate transcription initiation. Bacteria always contain one primary or so called “housekeeping” σ factor. In the gram-positive antibiotic producing bacterium, Streptomyces coelicolor, 65 genes encoding different σ factors are found. HrdB is the main sigma factor in S. coelicolor and is responsible for expression of essential genes. On the other hand, genes that are required in different stresses or environmental conditions are regulated by alternative σ factors. Alternative σ factors are typically about half the size (~30 kDa) of HrdB and lack some of its domains. These factors recognize promoter consensus sequences different from HrdB. They are responsible for transcription of gene subsets with specific functions1,2
