We used flow cytometry to detect the often-inconspicuous transitions among life cycle stages in three genera of chrysophyte algae (Chrysosphaerella, Ochromonas, and Synura). We revealed an alternation of two ploidy levels over time in strains maintained in long-term cultivation, both life cycle stages were capable of mitotic growth and shared similar phenotypes. Therefore, chrysophytes have a haploid-diploid life cycle with isomorphic life cycle stages. As far as we know, this is the first report of such life cycle strategy in unicellular algae. Life cycle stages and life stage transitions seem to be synchronized among all cells coexisting within a culture, possibly due to chemical signals. Additionally, ploidy level variation resulting from whole genome doubling was identified in two species of Synura.
