AnotaceGreen microalgae are among the most widely distributed microorganisms in the biosphere. They are significant contributors to global photosynthetic productivity and they are interesting for biotechnology area for their large variety of possible applications such as biofuels, cosmetics, pharmaceuticals, nutrition and food additives, aquaculture, and pollution prevention. Depending on the microalgae species, they can accumulate some compounds including a large range of chemicals and products, such as fats, polyunsaturated fatty acids, oil, natural dyes, sugars, pigments, antioxidants, high-value bioactive compounds [1]. Our aim is to understand the process of evolution and this knowledge can be used for higher compound accumulation/production of selected algae and cyanobacteria, for example higher production of astaxanthin by Haematococcus pluvialis or higher production of intracellular lipid bodies in Chlamydomonas reinhardtii. Combination several bio-imaging methods, signal processing techniques, high speed quantitative imaging cytometry, fluorescence activated cell sorting (FACS) and methods of molecular biology (highlighted characterization of whole genome and transcriptome analysis) we want to identify those candidates from populations which significant increase either in growth rate or compounds accumulation. Moreover, we would like to identify the evolutionary mechanisms related with the genome chages such as SNPs or futher mutations. Whole genome analysis is simplified by using powerfull method known as Next Generation Sequencing (NGS). We will focus on genome comparative analysis of our de novo breed strains and wild type strains so we enable to characterise evolutionary changes. These results will be useful for targeted engineering of new hyper productive strains that will increase the efficiency of biomass and other interesting compounds production in green algae.
