    AnotaceNanofibers are an attractive material, in which we can match their mechanical and chemical properties. Liver plays a major role in detoxification, protein synthesis, glycogen storage and many other functions. They form a complex structure composed from hepatocytes (60%), endothelial cells (10-20%), Kupfer cells (15-25%) and others. Mesenchymal stem cells (MSC) are multipotent stem cells isolated from the bone morrow and other tissues. They have significant immunomodulatory and trophic properties. Therefore we used MSC for co-cultivation with hepatocytes. In this study we created scaffolds for tissue engineering utilizing the methacrylate copolymer (HEMA-EOEMA) and poly-α-caprolactone (PCL) nanofibers as artificial extracellular matrix seeded with hepatocytes. We have assessed their viability and albumin production as an index of liver function. Moreover, the secretion of aspartate transaminase (AST) and alkaline liver phosphatase (ALP) have been measured in order to determine hepatocyte stress. In addition, co-cultures of hepatocytes and BMSC were maintained on nanofiber layers to evaluate the ability of MSC to support hepatocytes on nanofibers. The results show that the best carrier for cultivation of hepatocytes is PCL/EtAc/DMSO. Co-cultures of hepatocytes and BMSC increased the production of albumin and decreased secretion of stress markers within the time.
