In acentrosomal higher plants we found the presence of heterogeneous protein complexes of γ-tubulin in association with membranes and MTs. Large of them are active in microtubule nucleation. Inducible expression of RNAi constructs were used for γ-tubulin downregulation in Arabidopsis thaliana. Complete RNAi depletion of γ-tubulin led to the absence of microtubules. During partial RNAi depletion of γ tubulin, cells could progress through mitosis, but late mitotic events cytokinesis and phragmoplast formation were strongly affected, long anaphase spindles persisted between separated nuclei and polar distribution of γ tubulin during late mitosis was disturbed and often resulted in bi or multi-nuclear cells. The cell plate formation sites were often misaligned. Also in Arabidopsis seedlings development was observed multiple abnormalities. We suggested that some functions of γ tubulin that are important for cytokinesis, cell specification and polar growth might be microtubule independent.
