Techniques used at the Institute of Soil Biology ASCR for the cryopreservation of soil algae and cyanobacteria are introduced. Results from applying traditional 1 and 2-step freezing protocols, novel encapsulation-dehydration method, and different post-cryopreservation viability testing are presented. The advantage and limitations of particular procedures are discussed. Cyanobacteria were found to be the most cryopreservation tolerant surviving successfully direct plunging to liquid nitrogen without cryoprotectants. Among algae big differences were observed, several cryopreservation recalcitrant species were found. Encapsulation-dehydration method improved recovery of several freezing sensitive species and seems to be a promising method for cryopreservation of desiccation tolerant algae. A new NAJA imaging method has been developed to quantify post-cryopreservation recovery.
