Guanylate-binding proteins (GBPs) are components of cell-autonomous immunity against intracellular pathogens. They were shown to play an important role in response to multiple viral, bacterial, and protozoan pathogens, but before our research nothing was known about their role in leishmaniasis. In this invited comment we spotlight this novel mechanism of defense against Leishmania parasites that includes increase of expression of mRNA of GBPs after infection of different Leishmania species, involvement GBPs in direct killing of L. donovani in nonphagocytic cells, as well as their possible role in control of L. major in skin. We highlight the challenges and perspectives arising from this novel field of research.This invited comment-review summarizes the role of guanylate-binding proteins in leishmaniasis highlighting the challenges and perspectives arising from this novel field of research.
