Among nanovesicle drug delivery systems, liposomes are the oldest and most widely used
vehicles. To overcome some of their drawbacks, analogical supramolecular systems based on
different building blocks were developed in two recent decades. Next to polymersomes or
exosomes, an attention is turned to so-called dendrimersomes.3,4 Dendrimersomes are
nanovesicles most usually self-assembled from so-called Janus dendrimers. These dendrimers
consist of two different dendrons, where one bears lipophilic groups on its periphery and one
bears polar groups. Following dendrimersomes design, this project aims on preparation of
similar nanovesicles from amphiphilic molecules with one dendritic positively charged part and
one part formed by linear lipophilic alkyl chains as in the case of building blocks of liposomes. In the aqueous solution, these amphiphiles should be able to self-assemble into supramolecular objects. These supramolecular objects could then serve as nanovehicles for variety of drugs or nucleic acids with therapeutic effects. The project is specially focused on delivery of siRNA (short interfering RNA).
