High fluence lowpower (HFLP) lasers has been recognized as a promising tool for treatment of various pathological conditions. In addition, the precise molecular mechanisms of laser light on cells remain elusive. In addition, HFLP laser irradiation is widely used in superresolution localization microscopy. However, side effects of laser irradiation are largely ignored. A better understanding of the molecular mechanisms of HFLP lasercell interaction will be an instrumental for the development of novel therapeutic modalities. Therefore, we studied subcellular mechanisms involved in the laser interaction with human hepatic cell lines. We compared the effects of different laser wavelengths (398nm, 505nm, 650nm) on three various human liver cancer cell lines (Huh7, HepG2, Alexander). Our results revealed mitochondria as a subcellular “sensor” and “effector” of laser light nonspecific interactions with cells.
