Three-dimensional (3D) culture systems known as organoids derived from precursor cells are accessible for many tissues with a limited number of studies available for bone. The system allows the appropriate cell-cell interactions, however, achieving 3D spheroids from
the bone-related cells is still rare. Here we established a simple method for culture of 3D bone spheroids that can be used for bone research, translation medicine, disease modelling and regeneration. We also designed the prototype of a miniaturized culture chambre inside a Petri dish provided by continual perfusion of nutrients to minimalize a necrotic core formation and support osteogenic differentiation. The system also includes a live imaging system to observe the behaviour of cells.
