    The super-resolution (SR) methods are rapidly developing approaches in light microscopy field. The Stimulated emission depletion (STED) microscopy is SR method based on confocal scanning microscope. During the acquisition scanning, two lasers – excitation and depletion – are co-aligned into one single spot in the sample. For the maximal efficiency of the SR method, the proper co-alignment of used laser beams is crucial. The article explains the STED microscope operator recommended routine for the alignment. Simple experiments demonstrate significantly decreased number of incoming photons in the misaligned system. It naturally leads to the lower resolution during the acquisition and these important aspects are discussed within the article.
