Gadolinium-based MRI contrast agents recently raised concerns because of long-term gadolinium deposition in patients. It is known that kinetic inertness of the gadolinium chelates is to be blamed for release and retention of gadolinium. In this work, kinetic inertness of 46 macrocyclic gadolinium chelates was studied with two different methods. In general, chelates that were stable under acidic conditions were also stable under zinc transmetallation (neutral pH), but not vice versa. Coordinating acetate arm was found to contribute most strongly to kinetic inertness and many commonly used structural motifs were found to have detrimental effect.
