    The LINE-1 retrotransposon comprises 17% of the human genome, although only a few intact copies currently remain “hot”, capable of retrotransposition. LINE-1 elements have long been used as a cancer biomarker and, more recently, their expression has been associated with residual cancer stem cells resistant to chemotherapy. This poses a challenge as LINE-1 derepression can result in further cancer-driving mutations, both through somatic retrotransposition or through inter-LINE-1 recombination. FAM208A and other members of the HUSH (human silencing hub) complex have recently emerged as novel restriction factors for LINE-1 transposable elements. We will discuss these findings in context with previously known functions of the HUSH complex and discuss the prospectivity of targeting this complex for cancer treatment.
