Gadolinium-based MRI contrast agents release and deposit gadolinium in patients. Little is known about the relationship between structure of macrocyclic contrast agents and their ability to release gadolinium. This work shows that decreasing basicity of macrocycle nitrogen atoms and use of non-protonizable coordinating arms help prevent gadolinium release from the macrocyclic agents. These are promising ways for construction of safer gadolinium-based contrast agents.
