The whole-cell optical fiber sensor is proposed as a real in-situ detector for on-line monitoring of the environmental pollution in remote localities. This sensor needs an optimal design of an optical fiber element for ensure effective coupling of low intense light produced by bioluminescent bioreporters. We calculated that only 3.5% of all photons, produced by one bacterium placed on the front end of the optical fiber - polymer cladded silica fiber, are coupled and guided. Up to six times higher intensity of detected light was calculated by enlargement of an area covered with bacteria using optical fiber element with maximal transmittance. The results were confirmed by measurement with immobilized Pseudomonas fluorescens HK44. The influence of physical properties and optical fiber geometry, cell immobilization into silica gel or bacterial plaque, on the intensity of detected light, were evaluated.
