Functionality of a microfluidic device made of polydimethylsiloxane for testing epithelial-mesenchymal transition of cancer cells is presented. Arrays of micropillars as obstacles with different spacing are a prerequisite of high throughput measurements and standard evaluations of cellular deformability and migratory activity. The miniaturization of the device with the total migration path length less than 2 mm shortened analysis time by two orders of magnitude, when compared with commonly used devices. The device is compatible with multimodal holographic microscope in a time-lapse setup to visualize variability of mechanical properties of many individual cells under identical conditions. This arrangement enables observations unaffected by artifacts caused by phase contrast, due to a fluorescent staining or intensive light fluxes. Cells were subsequently monitored for several hours using holographic microscope Q-Phase (Tescan) with objectives Nikon Plan 10/0.30. Holograms were captured by CCD camera (MC-VELETA).
