This chapter summarizes the basic mechanisms of plant adaptation to drought stress and the use of plant phenotyping methods to detect plant drought tolerance. The mechanisms of plant adaptation include avoidance of plant water deficits, water deficit tolerance and the efficiency mechanisms. Specific exper-iments demonstrated the potential of using phenotyping methods to detect drought adaptation mecha-nisms. These include the following: i) root phenotyping, which allows the evaluation of differences in root system architecture and in particular, the analysis of root architecture traits associated with drought toler-ance; ii) infrared thermal imaging to evaluate stomatal responses to drought and water use efficiency, and iii) spectral reflectance, which enables the evaluation of the impact of drought on the change in the leaf structure and pigment composition. All three phenotyping methods tested represent potential tools for screening genotypes that are tolerant to drought. However, each method has some advantages as well as limitations and can be thus used to detect different mechanisms of drought tolerance.
