Glioblastoma multiforme is the most common, aggressive and chemorefraktory brain tumor in human adults. Microtubule-severing ATPases spastin, katanin and fidgetin are important regulators of microtubule organization. We have shown previously, that spastin is ovexpressed in gliomas and its depletion in human glioblastoma cell line T98G caused decrease of cell motility and increase in cell proliferation (J. Neuropathol. Exp. Neurol. 70:81, 2011). Here we report on expression, localization and function of katanin subunits p60 (catalytic) and p80 (regulatory) in human glioblastoma cell lines. RT-qPCR revealed differential expression of katanin subunits in glioblastoma cell lines. Both subunits localized to centrosomes, mitotic poles and leading edges of T98G cells. Free cell migration and cell morphology was changed in katanin-depleted T98G cells. We also observed changes in cell proliferation in katanin depleted cells. Collectively, our data indicates for the first time that katanin can regulate cell migration and proliferation in glioblastoma cells.
