Mast cells play an essential role in immune system. Their committed progenitors are released from the bone marrow to circulate in the bloodstream. Subsequently, they migrate into peripheral tissues to undergo terminal maturation. Chemoattractant-directed migration (chemotaxis) of mature mast cells or their progenitors is one of the key mechanisms responsible for their local accumulation. Better understanding of mast cell functions is conditioned by deeper understanding of molecular mechanisms of mast cell migration and the role of cytoskeleton in this process. Various methods for studying cell migration have been established, but they have limitations for exploring chemotaxis in mast cells. Here we report on a new technique based on real-time cell tracking that makes it possible to study migration of non adherent mast cells to chemoattractant. Presented method is a valuable tool for exploring chemotaxis of mast cells and cell migration in other cell types.
