    The lecture was about our recent data of the regulation of microtubule dynamics during mast cell activation. The key event in allergy response and inflammation during mast cell activation is change in the location of microtubules and their accumulation in the cell periphery. We found that mast cell activation stimulates generation of Ca2+-dependent microtubule protrustions. Calcium ions regulates generation of these protrusions through binding to calcium-binding messenger protein calmodulin. We also found that concentration of calcium ions as well as inhibition of calmodulin regulate microtubule nucleation from centrosome in nocodazole wash-out experiments. Moreover, enhanced levels of calcium modulate interactions of γ-tubulin with γ-tubulin complex proteins 2 and 4. The presented results revealed that Ca2+ concentration play an important role in the regulation of microtubule (+) end dynamics and microtubule nucleation in bone marrow mast cells.
