Wild-type and TLR2-/-, TLR4-/- and MyD88-/- deficient mouse bone marrow-derived dendritic cells were stimulated with formalin inactivated B. longum. BALB/c mice were monocolonized by bifidobacteria immediately after birth naturally from their gnotobiotic mothers in plastic isolator. Two-month old monocolonized mice and age-matched germ-free controls were three times subcutaneously immunized with major birch pollen allergen Bet v 1/Alum in two-week interval. Mice were sacrificed one week after the last immunization. Stimulation of murine dendritic cells by B. longum induced only low levels of co-stimulatory molecules CD40, CD80 and CD86 accompanied by production of IL-10, which was TLR2- and MyD88-depend. Bifidobacterium-colonized mice showed a significantly reduced level of allergen-specific antibody IgE, IgG2a, IgG1 and total IgE in blood sera compared to germ-free controls
