We studied the functional involvement of human YME1L in mitochondrial biogenesis and homeostasis using stable RNAi and expression experiments in HEK293 cells. We found that YME1L performs proteolytic control of mitochondrial OXPHOS biogenesis, functions antiapoptotic and is required for normal cell proliferation. Our results stress the importance of mitochondrial protein quality control for mitochondrial and cellular integrity and demonstrate novel mechanisms controlling respiratory chain biogenesis
