We show that cortical endoplasmic reticulum (ER) is involved in functional compartmentalization of plasma membrane as it confines the immediate communication between plasma membrane and cytosol to distinct (ER-free) zones. Our results further demonstrate that MCC/eisosomes are also involved in this process by regulating the distribution of cortical ER along the cell periphery.
