Classical methods of semen evaluation are based on three parameters: sperm concentration (x .106 ml), motility (%), and morphology. According to these parameters semen samples are classified as normozoospermic or asthenozoospermic, oligozoospermic, etc. Our long-term objective is to reveal causes that lead to sperm defects, i.e. to elucidate the significance of individual proteins for development of spermatozoa, that are able to fertilize the egg. Our essential approach is comparative analysis of normozoospermic sperm samples and samples with pathological parameters. For this purpose, we have first generated a set of monoclonal antibodies to human sperm proteins, which are successfully applied in evaluation of human semen in IVF laboratories. Second, we employ proteomic analysis to determine the sperm quality. Two-dimensional polyacrylamide gel electrophoresis (2D PAGE) and mass spectrometry (MS) are very promising approaches. They make possible: 1. identification of proteomic differences between normal and defective sperm 2. identification of numerous sperm-specific proteins 3. deeper understanding of protein functions in sperm motility, capacitation, acrosome reaction and egg fertilization
