Optical fiber biosensors based on monitoring of cell bioluminescence are employed as environmental pollution sensors. An active layer containing bioluminescent cells on the fiber element end face is a basal part of an optical fiber biosensor (OFB). The number of immobilized living cells and light coupling efficiency determine of OFB sensitivity. In this paper, two shapes of optical fiber elements (OFE) are compared; 600 μm polymer cladded silica (PCS) fiber, and tapered quartz element with 6.3 mm diameter on the front end narrowing to 800 μm (length 32 cm). On these ends, cells Pseudomonas putida TV A8 were immobilized in silica matrix. Bioluminescence measurements confirmed a proportionality between the number of cells and bioluminescence. The number of immobilized cells was 105 times higher on the tapered OFE end as compared to the 600 μm PCS fiber. The measured bioluminescence intensity increased 4 times in the tapered OFE case.
