From a complex lupulin gland-specific cDNA library from Osvald’s 72 hop, we isolated first hop-specific allelic forms of transcription factors (TFs) HlbHLH2 and HlWD40_1, which share high homology with well-described TT8 and TTG1 TFs from A.thaliana, respectively. These TFs are involved in combinatorial control of light-responsive and tissue-specific activation of phenylpropanoid pathway. HlbHLH2 and HlWD40_1 are quite lupulin gland-specific and, according to our transient expression experiments, they form specific complexes by interacting with previously isolated HlMyb2 and HlMyb3. The complex formation leads to strong activation of chs_H1 promoter. The activation is dependent on the presence of PMyb-like box CCWAC positioned in 5‘ end of the promoter. The interplay and regulation of expression of these crucial TF complexes could co-determine the rate of accumulation of valuable lupulin metabolites
