Intracellularly removable polymer nanocoatings designed for surface modification of siRNA-based polyplexes (PEC) were developed with the aim to improve properties of PEC in physiological environment. N-(2-hydroxypropyl)methacrylamide (HPMA) based copolymers containing reactive groups bound via biodegradable disulfide spacers to the polymeric backbone were synthesized and used for coating of PEC formed by the self-assembly of siRNA with the polycations polyethylene imine and poly(HPMA)-grafted poly(l-lysine) (GPL). Light scattering analysis showed the attachment of, on average, 28 polymer molecules to the surface of the polyplexes forming a 5-nm-thick hydrophilic stealth coating. Coating of the siRNA/GPL PEC substantially improved their stability in physiological solutions and an initial proof-of-principle for time- and concentration-dependent specific gene silencing was obtained. It was demonstrated that siRNA/GPL complexes hold significant potential for further development.
