Phenol-degrading strains Rhodococcus erythropolis CCM2595 and Rhodococcus jostii RHA1 were chosen for genetic analysis. We have cloned and sequenced the gene cluster (7 kb) responsible for phenol degradation in R. erythropolis strain: pheA1, pheA2 (subunits of phenol hydroxylase), catA (catechol 1,2-dioxygenase), catB (muconate cycloisomerase), catC (muconolactone isomerase), pheR (AraC-type regulator) and catR (IclR-type regulator). Genome sequence analysis of R. jostii RHA1 revealed presence of two similar clusters with the same arrangement of the phenol degradation genes. Using MALDI-TOF mass spectrometry, binding of the CRP-like protein upstream of the pheA2A1 genes of two Rhodococcus strains was proved. The CRP protein may be involved in regulation of substrate utilization hierarchy
