Antimicrobial drugs that target directly the cell surface structures, resulting in cell permeabilization, were screened by our fluorescence method based on the use of the potentiometric fluorescent probe diS-C3(3). DiS-C3(3) is a substrate for two major S. cerevisiae MDR pumps, Pdr5p and Snq2p. Its use represents an efficient strategy for identifying pump inhibitors with minimal side effects on membrane potential or membrane integrity, and for detecting cells permeabilized by surface active drugs. The screening is performed on cells in well-defined physiological state and does not require any pretreatment or preincubation
