Recognition and binding of spermatozoa to the zona pellucida (ZP) is a crucial step in the fertilization process. This is the topic of many scientific groups, and so far many molecules on the sperm surface, involved in mentioned process, were identified. Sperm surface proteins of ejaculated and capacitated boar sperms were isolated by Pierce® Cell Surface Isolation Kit and characterized by 1D, 2D electrophoreses, glycoprotein staining, hyaluronidase and proteolytic activities. Finally binding assay with ZP was performed. Results have shown the quantitative and qualitative differences in both 1D and 2D electrophoretic profiles, glycoprotein profile was similar to 1D electrophoretic one. Neither hyaluronidase nor proteolytic activities were observed. Binding assay proved that interactions occur between both ejaculated and capacitated sperms. Protein interactions of capacitated sperm with ZP glycoproteins seem to be more specific.
