Broken ends in three sex-chromosome mutant lines of the flour moth, Ephestia kuehniella , were examined by fluorescence in situ hybridization (FISH) with the insect telomere probe (TTAGG)n on spread preparations of pachytene oocytes. No hybridization signals were found at cut ends of Z-chromosome fragments and at one end of the deficient W chromosome. This implies that the broken ends had not been restored by a synthesis of telomeric DNA de novo or by another mechanism. In spite of the telomere loss, the broken ends of sex chromosomes evidently did not cause chromosome instability, cell cycle arrest or apoptosis. In contrast, they persisted in ASF lines for more than 50 generations (more than 11 years).
