Specimens of human adult bones from Žatec cemetery (11th-13th century) were investigated for determination of sex by molecular genetic methods. Sequences length difference in amelogenin gene locus on both X and Y chromosome was chosen for its value in sexing. A modification of the silica method for extraction of aDNA was used. In 94.7% (18 out of 19) of specimen extraction was successful. Obtained data were then confronted with the results given by morphological sexing methods and the concordance of both techniques was 83.3%. This observation can be also taken as an indirect proof of authenticity of extracted aDNA fragments.
