The development of miniaturized liquid chromatography systems (LC) for in situ analysis requires adequate miniaturization at the level of individual system components. An important component of any LC system is the detector, which is responsible for detection of the separated components and generating a signal that can be used for identification and quantification of analyte. In optical detectors, the design of the detection flow cell plays a crucial role in the overall performance of the LC system. Herein we present some preliminary results characterizing compact UV-Vis absorbance detector comprising miniature deuterium lamp, 97 nL capillary flow cell and CCD spectrometer. Detection cell is coupled to the light source and CCD spectrometer by the optical fibres integrated in the flow cell structure. In the cell performance test, acetone-water mixture was measured and the effective optical path of the cell was 5.3 mm. Absorbance baseline noise depended on the selected wavelength (210 – 610 nm) as well as on the integration time (25 – 200 ms) and was in the range of 1.332 – 9.405 mAU. In isocratic chromatography separation of the test mixtures, the performance of the proposed detector was compared with the commercial DAD detector equipped with 80 nL rectangular flow cell. In conclusion, the proposed detector and capillary flow cell design are suitable for portable LC instruments.
