Polymeric conjugates based on N-(2-hydroxypropyl)methacrylamide (HPMA) containing doxorubicin bound via an amidic bound directly penetrate the plasma membrane and are detectable in all associated cellular membranes. We have never been able to detect released doxorubicin inside the nuclei of the treated cells. Necrosis is the main mechanism of the cell death. Conjugates containing hydrolytically bound doxorubicin are internalized by endocytosis and fluid phase pinocytosis and doxorubicin is cleaved from th polymeric carrier at low pH in late endosomes and lysosomes. An apoptosis is the main mechanism of the cell death
Polymerní konjugáty na bázi N-(2-hydroxypropyl)methacrylamidu (HMPA) nesoucí doxorubicin navázaný amidickou vazbou, procházejí přes plazmatickou membránu a jsou akumulovány ve všech souvisejících buněčných membránách. Nikdy se nám nepodařilo prokázat odštěpený doxorubicin v jádrech buněk. Hlavním mechanismem buněčné smrti je nekróza. Konjugáty nesoucí doxorubicin navázaný hydrazonovu vazbou jsou internalizovány endocytózou a pinocytózou a doxorubicin je odštěpen nízkým pH v pozdních endozómech a lysozómech. Hlavním mechanismem buněčné smrti je apoptóza
