IDEIS – identification of isoforms.

Technical parameters: IDEIS je soubor skriptů napsán v jazyce Python a R. Je určen pro single-cell RNA data genové exprese získaných sekvenací knihoven generovaných za pomoci 10X Chromium Single Cell 5’ Kitů. Ze seznamu exonů a pravidel na konstrukci umelých “transkriptů” skripty vyhotoví co nejvhodnejší “transkriptom” pro určení isoforem CD45/PTPRC, na který jsou pak mapovány ready za využití externího programu Salmon Alevin. Data jsou pak zpracovány v R pro co nejsnadnější manipulaci pri dalších analýzách. Tento software umožňuje analýzu isoforem bez nutnosti další úpravy experimentu. Economic parameters: Úspora nákladů na sekvenačním kitu a protilátkach v hodnotě 20 000 Kč na experiment. Úspora času při přípravě označování buněk protilátkami během přípravy single-cell experimentů nevyžadujících jiné protilátky a jejich následné analýzy v hodnotě cca. 20 človekohodin na experiment.
