The principal properties of SPBs are mentioned together with their use as geometrically well-defined models of biological membranes. The basics of each of techniques are explained and their possibilities and limitations are discussed. Images with diffraction-limited resolution are obtained by CFM showing the spatial distribution of fluorescent molecules in the sample. Selective labeling offers the possibility to visualize location of different molecular species. Furthermore, the signal from each point of the sample can be analyzed separately; the measurement of molecular mobility in individual points of the sample by fluorescence correlation spectroscopy is addressed in more detail.
