We studied genetic regulation of IgE level in humans and its impact on development of allergic diseases using information on previously reported IgE-controlling locus in mice to test their orthologous region for regulation of total IgE and numbers of specific IgEs in humans (Czech atopic patients). A new IgE-controlling locus was identified on human chromosome 8q12 that influences sensitization to a number of allergens. The finding of this linkage shows the precision and predictive power of mouse models in investigation of complex traits in humans. To define other IgE-controlling loci in the patients we selected markers in further 16 candidate regions for linkage and association testing with atopic phenotypes. By candidate gene approach we confirmed the role of previously reported loci 5q33.3, 7p14.1, 12q13, 13q14 in IgE control and development of atopy.
