Její princip využívá skutečnosti, že dielektrická částice s indexem lomu vyšším než je okolní médium, je tažena do místa s největší intenzitou laserového svazku. Tyto síly se pohybují v řádech jednotek až stovek pN a umožňují prostorové zachycení dielektrických objektů o velikostech od několika desítek nanometrů po desítky mikrometrů. Doplní-li se sestava citlivým systémem pro detekci výchylky zachyceného objektu a vhodnou kalibrační metodou, lze ji využít jako citlivý profilometr pro měření povrchů, ke kterým není přímý mechanický přístup a které jsou např. v imerzním prostředí. Description in English In recent years, optical tweezers have become an invaluable tool at study of power interactions between micro-particles, nano-particles, live cells, sub-cellular structures and individual macromolecules [1,2]. Their princi ple exploits the fact, that a dielectric particle with the refraction index higher than the surrounding medium, is pulled to the place with the biggest intensity of the laser beam. These powers span in orders from units to hundreds of pN and enable spatial trapping of dielectric objects of sizes from several tens of nanometers to tens of micrometers. If the configuration is supplemented with a sensitive system for detection of deviation of the trapped object and a suitable callibration method, it is possible to use it as a sensitive profilometer for measurement of surfaces, which are not directly mechanically accessible and which are e.g. in an immersion environment.
