We investigated cell lysosomalmediated signaling in three hepatic cell lines (Huh7, HepG2 and Alexander) over magnetic nanoparticles treatment in different concentrations. Nanoparticle treatment resulted in reorganization of Factin and tubulin cytoskeletal filaments. Lysosomal accumulation of nanoparticles induced significant lysosomal dysfunction and affected mTOR activity. Further, we found that nanoparticle treatment triggered autophagic flux in Alexander and Huh7 cells, whereas HepG2 remained unresponsive to the treatment. Indeed, Bcl2 protein expression level was a key to the resistance of HepG2 cells to nanoparticle treatment.
