Multimodal imaging probes that combine different metals within a single molecule are difficult to synthesize because the position of the individual metals is hard to control. This work presents a new amino acid chelator building block (DO3A-Hyp) that allows incorporation of metals into peptides during the peptide synthesis. With this approach, it is possible to tightly control the positions of the metals within the peptide molecule. Peptides containing two different metals with known positions were successfully prepared, paving the way for efficient preparation of multimodal probes or theranostic agents.
