The topic of this paper was modulation of expression of the genes involved in the biosynthesis of branched-chain amino acids in Corynebacterium glutamicum. The activity of the promoters P-ilvD (dihydroxyacid dehydratase) and P-ilvE (transaminase B) was up-modulated and the activity of the promoters P-ilvA (threonine deaminase) and P-leuA (isopropylmalate synthase) was down-modulated by site-directed mutagenesis. The previously constructed strain C. glutamicum ilvNM13 with acetohydroxy acid synthase, resistant to inhibition by all three branched-chain amino acids (L-valine, L-isoleucine and L-leucine), was used as a basis to develop a new type of valine producer by genetic engineering. A constructed weak promoter of ilvA (or leuA), which was introduced into the C. glutamicum chromosome via a gene-replacement technique reduced the biosynthetic rate of isoleucine (or leucine), which lowered the mutant growth rate and increased valine production
Tématem tohoto článku byla modulace exprese genů zúčastněných v biosyntéze větvených aminokyselin v Corynebacterium glutamicum. Aktivita promotorů P-ilvD (dehydratasa dihydroxykyselin) a P-ilvE (transaminasa B) byla zvýšena a aktivita promotorů P-ilvA (threonindeaminasa) a P-leuA (isopropylmalátsyntasa) byla snížena místně-specifickou mutagenezí. Dříve vytvořený kmen C. glutamicum ilvNM13 se syntasou acetohydroxykyselin, rezistentní k inhibici zpětnou vazbou všech tří větvených aminokyselin (L-valinu, L-isoleucinu a L-leucinu), byl použit jako základ pro vývoj valinového producenta nového typu genovým inženýrstvím. Zkonstruovaný slabý promotor genu ilvA (leuA), vnesený do chromosomu C. glutamicum technikou výměny genů, způsobil omezení biosyntézu isoleucinu (leucinu), které se u mutant projevila snížením růstové rychlosti a zvýšením produkce valinu
