Sulfated carbohydrates are components of many glycoconjugates, and are degraded by two major processes: cleavage of the sulfate ester by a sulfatase, or en bloc removal of a sulfated monosaccharide by a glycoside hydrolase. We describe here the synthesis of a series of p-nitrophenyl b-D-glucosaminides bearing sulfate esters at the 2-, 3-, 4- or 6- positions, by divergent routes starting with p-nitrophenyl 2-acetamido-2-deoxy-b-D-glucopyranoside. The sulfated p-nitrophenyl b-D-glucosaminides were used to study the substrate specificity of four sulfatases, and revealed significant differences in the prefence of each of these enzymes for desulfation at different positions around the sugar ring

Sulfatované sacharidy jsou součástí mnoha glykokonjugátů a jsou odbourávány dvěma způsoby: odštěpení sulfátu sulfatasou nebo odstranění celého sulfatovaného monosacharidu glykosidasou. V této práci popisujeme přípravu sady p-nitrofenyl-b-D-glukosaminidů sulfatovaných v pozicích 2, 3, 4 a 6 na svém cukerném kruhu syntézou vycházející z p-nitrofenyl-2-acetamido-2-deoxy-b-D-glucopyranosidu. Připravené látky byly použity jako substráty pro čtyři sulfatasy a 24 fungálních b-N-acetylhexosaminidas pro zjištění variability substrátové specifity těchto enzymů
