Language eng - English Country US - United States Keywords cumulus cells ; extracellular matrix ; hyaluronan Subject RIV EB - Genetics ; Molecular Biology R&D Projects GA305/05/0960 GA ČR - Czech Science Foundation (CSF) CEZ AV0Z50450515 - UZFG-Y (2005-2011) Annotation Our results, together with the high sequence similarity found among human, rabbit, mouse, rat, bovine and equine TNFAIP6 and the expression of this protein and/or the respective gene in preovulatory follicles of all the examined species, strongly support the concept that TNFAIP6-mediated HC covalent transfer onto hyaluronic acid (HA) synthesized by cumulus cells is a mechanism common to mammalian OCCs. However, in contrast to the situation observed in the mouse, anti-TNFAIP6 A38 antibody did not prevent cumulus expansion nor HA accumulation in the matrix of pig OCCs stimulated with FSH. It suggests that porcine OCCs have evolved supplemental mechanisms for holding HA in the matrix. Thus, it is reasonable to hypothesize that different relative levels of HA, TNFAIP6, versican as well as other matrix components are synthesized by cumulus cells of different species, with the aim to create for each species the matrix that is the most suitable for ovulation and oocyte fertilization.
