Site directed mutagenesis was used for introduction of point mutations in the H4-H5 loop of Na+/K+-ATPase. Influence of these mutations on the enzymes function was evaluated by fluorescence probe TNP-ATP and its competitive replacement by ATP. Eight amino acids were identified to form the ATP-binding site.
